I have a pair of Sonos One speakers purchased in 2019. I connect to a desktop computer to Spotify and sometimes from my mobile device. I need to add another pair of Era 100 speakers to a different room. So they will be on the same wifi network. Is it possible to connect one device to one pair and a different device to the 2nd pair while both Sonos pair are on the same wifi network?

Each controller is not limited to connecting to only one set of speakers, but the system as a whole.
The Ones and Era 100s will both show on the desktop app and your mobile app as available rooms.
You will have the ability to control each room from both your desktop and mobile.
Not sure what listening to Spotify on your desktop has to do with Sonos?
You can have a number of Sonos.devices/speakers and play different music on each.
If you have them paired or bonded into Sonos Rooms then you can play different music on each Room.
Thank you! On Spotify, I was just providing info on how I use the 1 set I have now. Spotify runs on my desktop and my desktop is the sound source for the Sonos, except sometimes when my kids connect to them on their mobile devices. May not be describing it exactly right, tech really isn't my thing.

Do you mean you are running the Sonos Controller program on your computer?
The Controller is just that, like the remote control of your TV set, no music passes through it or your computer, the Sonos devices connect to the music source directly at the command of the Controller.

Yes, you can connect different devices to different Sonos speaker pairs on the same WiFi network and stream to each pair independently without any issues.

The takeaway for you is that the controllers act independently. All controllers can operate all Rooms. Rooms can be Grouped to play the same music. Unless the music is physically stored on a particular pad/phone, play can be initiated by any controller.
In the case of families, it may take a few days for each party to work out the personal discipline required to always operate the most appropriate Room. In case of pranks, I suggest that this can work both ways and a truce will soon follow.
