When setup as a stereo pair, yes. Whether or not you hear different instruments and vocals out of different speakers depends on the source material and how it was recorded.
Well recorded stereo sound is more than one instrument from one and the other from the other speaker - that often leaves a hole in the middle. What well placed stereo speakers - including Sonos - will do with good stereo recordings is to present a stage of the sound/music in front of you, including that appearing to come from between the two speakers.
I've found that I really prefer stereo pairs of Play 1s or Play 3s to just mono from one, it sounds kind of flat to me and I get tired of it fairly quickly. I do have mono speakers in the bath and garage where getting stereo set up would be difficult.
The Play 5 sounds good enough to me that I don't feel a need to pair them.
