Will a sub gen 2 work with gen 1 play 1,3 & 5 as this is my current set up?

Yes, it will work with those speakers, but you can only choose one of those speakers to add the Sub to. You can still group the speakers together as always.
FYI… The only products the Sub cannot work with is the Connect, Port, Move, and Roam.
