I have 4 Zoneplayer Zp100's, one connected to the router by ethernet cable, a playbar and a room with two PLAY:5's. The audio can drop out once in a while in the PLAY"5 room and it is also the weakest wifi area of my house. Could a BOOST be strategically placed somewhere between the rooter and the room in question to enhance the SONOS connectivity there or was it designed only to be an device to connect directly to a router?

Yes, it would, and no it isn’t. 😉
Although for complete transparency, you could also put another speaker between the two locations, and it would do the same thing, with the benefit of adding more sound and music to your home. It doesn’t have to be a BOOST.
I'd be tempted to look at adding a Play 1 for not much more than the Boost too.
