Hi I will arrive tomorrow the sub Mini. I have Beam 2 and a pair of Era 100 setting as sorround.
once I connect the mini sub do I have to redo the trueplay or can I leave the one I did 5 days ago when I activated the Era 100?

You will need to re-do the Trueplay tuning of the HT setup again after adding the Sub-Mini.
