Is it possible to set it up to mute both microphones on a paired set of Sonos Ones by pressing the mute button on just one of the pair?

I don't have the Ones so I don't know but I'm guessing not
No. The button requires a physical push on each speaker to Mute or Unmute the mic. It is designed this way so that it cannot be changed remotely and impact on your privacy. This is a good thing.
