I have a small HTS consisting of a Beam, and two One SL’s, and I am about to get a Port to connect my turntable to the system. My question is can you isolate the two SL’s to play the audio from the turntable as a group, then automatically returning to “TV Mode” ie including the Beam back into the group when the TV is turned back on?

You can’t easily isolate the One's from you surround system. You can set them up as a stereo pair and then groeup them to your Port, but to use them as surrounds you’d have do the surround set up again (including Trueplay).

You should get decent sound from the turntable via the HT set up from the Port if you set the HT to music mode such that the One units become full participants in music play, in addition to the Beam.

And consider buying a Play:5 instead of a Port. A bit more money but a lot more product.

The Port is a very expensive way just to add a line in.
