I have a large room and was wondering if I could put 2 lots (4 play:1s) as stereo pairs

Yes, you can. Or play four units as independent speakers but as one grouped zone. Or as one stereo pair and two independent speakers, as a group. Try the combinations and settle on what works best for you/your space. With Sonos it is easy to chop and change to see what suits best.
